Dr. Nizetic was one of the first scientists who cloned single, isolated human chromosomes in the form of genomic DNA libraries which were gridded and permanently stored, the well known ICRF-reference library system, which helped mapping many individual chromosomes DNA-s in the human genome project.
In 1994, Dr. Nizetic founded his independent research group. The group made a major contribution towards the final molecular map of the human chromosome 21, which culminated in the inclusion as a co-author of the historic paper on May 18th, 2000, in which the complete DNA sequence of human chromosome 21 has been published. This was only the second human chromosome to be completely sequenced.
Prof. Nizetic focussed on detection and characterization of genes from human chromosome 21, and their dose effects causing Down syndrome with all its interesting phenotypic features, such as childhood leukaemia, neuronal development abnormalities and Alzheimer’s disease.
In collaboration with Prof. K. Pavelic and his team at the “Rudjer Boskovic Institute” and with the use of the Croatian Tumour Bank, sponsored by the Croatian Academy of Sciences, the group of Prof. Nizetic cloned a novel gene, a ubiquitin specific protease, from the minimal region of deletions in lung cancer.
In 2003, Prof. Nizetic’s group was the first to discover that the acquired mutations in the transcription factor GATA1 occur in utero, and lead to the transient myeloproliferative disorder, but do not predict the onset or the prognosis of the ensuing acute megakaryoblastic leukaemia in Down syndrome children. Recently, Prof. Nizetic’s group completed the global transcriptome analysis using Affymetrix microarray (Gene Chip System). A new marker was discovered, specific for the progressive type of the leukaemic cells. This marker molecule has the potential to be exploited for therapeutic purposes. These results were published recently in Lancet and Brit. J. Haematology.
Recently, a transchromosomal embryonic stem cell model project to map the gene-dose effects of trisomy 21 was started by Prof. Nizetic’s group.
